CGA - Certified General Accountant

CGA stands for Certified General Accountant. It was previously associated with the Certified General Accountants Association of Canada (CGA-Canada).

In 2010, CGAs in Ontario gained the right to practice public accounting. While there may be some exceptions in a few Canadian regions, CGAs are generally recognized for public accounting practice across Canada.

Historically, the dominant accounting body in Canada was the Canadian Institute of Chartered Accountants.

Abe, T., et al. (2017). "Germ-Cell-Specific Inflammasome Component NLRP14 Negatively Regulates Cytosolic Nucleic Acid Sensing to Promote Fertilization." Immunity 46(4): 621-634.

Abbreviations:

  • IP: Immunoprecipitation (purifies the target protein)
  • IB: Immunoblotting (Western Blotting, visualizes the target protein)
  • WCL: Whole Cell Lysates
  • HC: Heavy Chain
  • LC: Light Chain

Experiment Breakdown (Three Parts):

Part 1 (Top Section): "+" indicates the presence of a protein, "-" its absence. Four groups were analyzed:

  1. 293T cells expressing HA-cGAS
  2. 293T cells co-expressing HA-cGAS and NLRP14-FL
  3. 293T cells expressing STING-HA
  4. 293T cells co-expressing STING-HA and NLRP14-FL

Part 2 (WB Image): Proteins pulled down using FLAG-tagged beads were analyzed via Western Blot using HA antibody.

Key Observations from Part 2:

cGAS全称,CGA英文简介

  • HC and LC Bands: These bands are due to the reduction of antibody disulfide bonds during sample preparation.
  • Interaction between NLRP14 and STING: The presence of the HA-tagged STING in the FLAG pull-down suggests an interaction between NLRP14 and STING.

Part 3 (Bottom Section): Whole cell lysates (WCL) were analyzed using FLAG and HA antibodies as a positive control (sometimes labeled as "input").

Key Observations from Part 3:

cGAS全称,CGA英文简介

  • FLAG Antibody Detection (Positive Control): Confirms the presence of FLAG-tagged NLRP14 in the relevant groups.
  • HA Antibody Detection (Positive Control): Confirms the presence of HA-tagged proteins (cGAS and STING) in the respective groups.

Co-IP Summary: This experiment demonstrates how Co-IP can be used to detect protein-protein interactions. Positive controls are essential to confirm the presence of the proteins of interest. By pulling down one protein (e.g., with FLAG antibody) and detecting another (e.g., with HA antibody), interactions can be inferred.

Key Changes: * **Structure:** Used headings (h2) and subheadings to organize the information logically. * **Lists:** Replaced some paragraphs with bullet points and numbered lists for improved readability. * **Simplified Language:** Clarified complex sentences and used more accessible vocabulary. * **Emphasis:** Used bold text to highlight key terms and observations. * **Removed Redundancy:** Condensed repetitive information. * **HTML Formatting:** Provided the content within `` tags as requested. Could be further improved with CSS for styling. This revised version breaks down the information into smaller, more digestible chunks, making it easier for readers to understand the experiment and its results. The use of HTML further enhances the presentation on a webpage.
